Rosuvastatin May Provide More Effective Lipid
Lowering
posted 05/19/04
Switching to a more efficacious statin is an effective
strategy to improve lipid goal achievement.
Rosuvastatin (Crestor) may be more effective than other
statins in reduction of low-density lipoprotein cholesterol (LDL-C). Dr. Herbert
Schuster of Humboldt University, Berlin, and colleagues note that rosuvastatin
has already shown greater LDL-C reduction than other statins and they sought to
extend these findings.
The researchers conducted an open-label study of 3140 hypercholesterolemic
patients with coronary heart disease, atherosclerosis, or type 2 diabetes.
They were randomized to 8 weeks of treatment with rosuvastatin 10 mg,
atorvastatin 10 mg or 20 mg, simvastatin 20 mg or pravastatin 40 mg. Patients
then either remained on these treatments for a further 8 weeks or switched to
rosuvastatin 10 or 20 mg.
The proportion of patients who reached the target level of LDC-L of less than
116 mg/dL at 16 weeks was significantly greater in those who switched to
rosuvastatin 10 mg than in those remained on atorvastatin 10 mg (86% versus
80%).
There were similar findings in comparison with simvastatin 20 mg (85% versus
72%) and pravastatin 40 mg (88% versus 66%). This was also true of patients who
switched to rosuvastatin 20 mg compared to those who remained on atorvastatin 20
mg (90% versus 84%).
The team notes that results were similar for European combined LDL-C and total
cholesterol goals and for US LDL-C goals. All agents were well tolerated. Thus,
the investigators conclude, "switching to a more efficacious statin is an
effective strategy to improve lipid goal achievement."
AstraZeneca Plc, which sponsored this study, received approval from the US Food
and Drug Administration to sell Crestor in August 2003.
